Overview

This product is a ready-to-use tissue RNA stabilization solution, which is non-toxic and can rapidly penetrate tissues. After samples are fully immersed in this solution, RNA integrity can be maintained under non-frozen conditions. It is suitable for the collection and transportation of various fresh tissue and cell samples, and enables long-term preservation of RNA samples at low temperatures.

Applicable Sample Range:

⁕ Animal tissues: liver, brain, heart, spleen, lung, kidney, muscle, skin, bone tissue, etc.

⁕ Cell samples: adherent cells and suspension cells.

⁕ Microorganisms: bacteria, fungi, yeast and so on.

⁕ Others: isolated leukocytes/lymphocytes, dewaxed plant tissues.

Instructions for Use

I. Sample Preparation and Treatment

1. Animal tissues: Only use fresh tissues. Cut tissues into small pieces with the maximum thickness no more than 0.5 cm in any dimension. Note that oversized tissue pieces will slow down the penetration of the preservation solution, which cannot inhibit RNase degradation effectively and eventually cause RNA degradation in the tissue core.

2. Cultured cells: Harvest cells by centrifugation and discard the culture medium; or directly add the stabilization solution to adherent cells.

3. Plant tissues: Remove the epidermis or chop up tissues if there is waxy or cuticular layer on the surface of leaves and stems to facilitate solution penetration.

4. Blood samples: Whole blood is forbidden for direct preservation. Separate blood cells such as leukocytes by centrifugation first, discard red blood cells and plasma, and only preserve cell pellets.

II. Addition of Stabilization Solution

1. Add the solution at a weight-to-volume ratio of sample to preservation solution from 1:5 to 1:10. For instance, add 0.5–1 mL preservation solution for 100 mg tissues.

2. Ensure all samples are completely immersed in the solution.

III. Storage and Transportation

Tightly cap the sample tubes, then transport at room temperature or store in refrigerator as required.

IV. Subsequent RNA Extraction

1. Take out tissues from the stabilization solution with tweezers without rinsing.

2. Directly add Trizol or lysis buffer for grinding and homogenization, and follow the protocols of RNA extraction kits for subsequent operations.

Frequently Asked Questions:

1. Precipitates appear in the preservation solution at low temperature. Is it still usable?

Answer: Yes. Crystal precipitation at low temperature is a normal physical phenomenon. Warm it up to room temperature before use, shake well to fully dissolve, then it can be used normally.

2. Why is the RNA extraction result unsatisfactory?

Answer: The most common cause is oversized tissue pieces. Be sure to cut tissues into pieces thinner than 0.5 cm, and ensure the volume of preservation solution is at least 5 times that of the tissue. Tissue freshness also affects preservation effect. Stale tissues contain high levels of RNase, which may have already caused RNA degradation. You can improve the effect by increasing the dosage of stabilization solution.
