Overview

Trypsin selectively hydrolyzes peptide bonds formed by the carboxyl groups of lysine or arginine in denatured proteins, making it an important digestive enzyme. Additionally, trypsin is widely used as an adjunct therapy for conditions such as local edema, hematomas, and abscesses resulting from empyema, hemothorax, surgical inflammation, ulcers, traumatic injuries, and similar cases. In this assay kit, trypsin catalyzes the hydrolysis of the ester bond in TAME, releasing free carboxyl groups. These groups undergo a neutralization reaction with sodium hydroxide present in the reaction system, leading to a decrease in the pH of the solution. Using phenol red as an indicator, the change in absorbance at 555 nm is measured, enabling rapid determination of trypsin activity. Within a certain range, trypsin activity exhibits a well-defined linear relationship with the decrease in absorbance at 555 nm.
