MS-grade Trypsin-N protease specifically cleaves peptide bonds at the N-terminus of lysine (K) and arginine (R) residues. It is a new series of proteases with N-terminal cleavage specificity. Physical Form: MS-grade Trypsin-N protease is supplied as a lyophilized powder, containing 0.47 mg HEPES and 0.05 mg CaCl₂ per vial. Reconstitution Solution: Reconstitute with 40 μL of 50 mM zinc acetate solution before use. pH Range: Trypsin-N exhibits optimal activity at pH 7.5. Application: Trypsin-N protease specifically cleaves peptide bonds at lysine (K) and arginine (R) residues in denatured proteins. General In-Solution Digestion Protocol: Recommended Digestion Buffer: 20–50 mM HEPES or Tris buffer. Reaction Conditions: Use a protein-to-protease ratio (w/w) of 20:1. Incubate at 37°C for at least 4 hours. Tips: 1. Maximizing Activity: It is recommended that protein samples undergo denaturation and alkylation, and that the digestion reaction system and SOP are strictly followed. 2. Sensitivity: Trypsin-N is sensitive to 1 mM DTT, 0.1 mM guanidine hydrochloride, 0.8 M urea, and 5% acetonitrile, which will reduce digestion efficiency.
