Overview
W5 Solution is a classic buffer system for the preparation, washing, and resuspension of plant protoplasts, formulated with sodium chloride, calcium chloride, potassium chloride, and MES. This system maintains appropriate osmotic pressure and ionic environment for washing, purification, and short-term preservation of protoplasts after enzymatic digestion, reducing the risk of protoplast rupture and maintaining cell membrane stability. It can also be used for equilibration prior to PEG-mediated protoplast transformation.
W5 Solution provides a mild osmotic environment, effectively removes residual impurities from enzymatic digestion, and minimizes interference from disrupted cell debris. It is widely used in plant cell biology experiments, including protoplast isolation, transfection, and fusion in species such as Arabidopsis thaliana and tobacco.
Note: Perform all operations under sterile conditions to avoid microbial contamination. Do not use if the solution appears turbid or contains precipitate.
