This staining solution is based on the classic Karnovsky-Roots ferrocyanide-copper method for the localization of acetylcholinesterase (AChE) activity in tissue sections. Cholinesterases (ChE) are classified into two types: Acetylcholinesterase (AChE, true cholinesterase), which is primarily distributed in neuronal cytoplasm and at neuromuscular junctions (motor endplates), where it hydrolyzes acetylcholine and plays a key role in physiological regulation; and Pseudocholinesterase (PsChE, butyrylcholinesterase), which is mainly found in plasma, pancreas, and salivary glands, though its physiological function remains unclear. Staining Principle: Acetylcholinesterase hydrolyzes the substrate acetylthiocholine iodide, releasing acetic acid and thiocholine. The thiol group (-SH) of thiocholine reduces potassium ferricyanide to potassium ferrocyanide, which then combines with copper ions to form an insoluble reddish-brown to dark brown cupric ferrocyanide precipitate at the enzyme active site, thereby precisely localizing enzyme activity. Features and Applications: This method is simple to perform and exhibits minimal enzyme diffusion artifacts; however, substrate penetration into tissues is relatively limited. It is suitable for observing the morphology and enzyme activity of central and peripheral nerve fibers in frozen sections. At the research level, it can be applied to histological studies of Hirschsprung's disease and intestinal neuronal dysplasia. In samples from organophosphate poisoning, acetylcholinesterase activity is inhibited, resulting in negative staining. Positive staining appears as reddish-brown to dark brown precipitates. Note: This product contains heavy metal salts and is an irritant. Wear gloves and safety goggles throughout the entire procedure, and avoid skin and mucous membrane contact.
