The NHS ester moiety in AF647-NHS ester (5C-Linker) is a commonly used amine-reactive group. It activates the carboxyl group on the AF647 molecule, enabling it to react with amino groups (primarily primary amines) on target biomolecules such as proteins, antibodies and peptides. This reaction forms stable amide bonds to visualize and label biomolecules. It is widely applied in flow cytometry, confocal microscopy and fluorescence-based assays for labeling and detection of cells, proteins and other biological entities. It can also serve as a sensitive reporter group in immunoassays. Ex/Em(nm)=651/671.

Precautions for Fluorescent Products:
1. Activated esters are prone to hydrolysis. After taking out from the refrigerator, allow the vial to warm to room temperature in a dry environment before opening. Since the reagent quantity is small, gently tap the vial several times before opening to ensure all powder falls to the bottom.
2. It is recommended to prepare the stock solution fresh at first use. Use the dissolved reagent as soon as possible to guarantee experimental performance.
3. Fresh anhydrous DMSO is required for dissolution. Water contamination will cause hydrolysis of the activated ester and compromise experimental results.
4. The prepared stock solution is susceptible to hydrolysis upon exposure to water. If not fully consumed in one experiment, aliquot the solution (e.g., 5–8 μL per tube), seal with parafilm, wrap with aluminum foil, place in a well-sealed plastic bag containing desiccant, and store hermetically at ≤ -20°C protected from light.
5. It is recommended to optimize cell density, dye/probe concentration and incubation time in preliminary tests before formal experiments to determine optimal experimental conditions.
