Plasma total cholesterol consists of two-thirds cholesterol esters and one-third free cholesterol. It is a key component of blood lipids, and both forms are closely associated with cardiovascular and cerebrovascular diseases as well as various pathological conditions. This kit employs the total cholesterol and free cholesterol detection methods recommended by the World Health Organization (WHO), the U.S. Food and Drug Administration (FDA), and China's National Clinical Laboratory Operating Procedures, offering high sensitivity with a detection range of 20–5000 µmol/L, as well as excellent reliability and reproducibility.
The kit utilizes the CHOD-PAP endpoint method combined with the classic GPO Trinder enzymatic reaction. The principle is as follows: (1) Cholesterol esterase hydrolyzes cholesterol esters into free cholesterol; (2) Cholesterol oxidase oxidizes free cholesterol to produce hydrogen peroxide; (3) Under the catalysis of peroxidase, the chromogenic substrate is converted into benzoquinone imine, with the optical density being proportional to the cholesterol concentration; (4) The total cholesterol concentration and free cholesterol concentration are calculated separately, and the difference between the two values represents the cholesterol ester concentration.
