Citrate synthase (CS, EC 2.3.3.1) is widely distributed in the mitochondrial matrix of animals, plants, microorganisms, and cultured cells. It is the first rate-limiting enzyme of the tricarboxylic acid (TCA) cycle and one of the major regulatory sites of this cycle. The assay principle of this kit is as follows: CS catalyzes the condensation of acetyl-CoA and oxaloacetate to form citryl-CoA, which is subsequently hydrolyzed to produce citrate. Concurrently, this reaction drives the conversion of colorless DTNB to yellow TNB, which exhibits a characteristic absorption peak at 412 nm. By measuring the rate of increase in absorbance at 412 nm, the enzyme activity of CS can be calculated.
