Cesarean section in aseptic mice

Summary

Aseptic mouse cesarean section, and artificial rearing are known as the two key technologies for the preparation of sterile mice from scratch. There are 2 main methods of cesarean delivery in sterile mice: aseptic uterotomy and aseptic hysterectomy.

Principle

Cesarean section of sterile mice, which is a mandatory way to culture sterile mice, is suitable for areas where it is difficult to obtain the original germline of sterile mice. The basic principle is that a healthy animal on the verge of giving birth is treated with anesthetic drugs and then cesarean section is performed according to aseptic surgery.

Materials and Instruments

Equipment:
Surgical scissors, hemostatic forceps, traction cord, sterile ferry, sterile gauze, mouse box, feeding isolator.
Reagents:
① povidone iodine;
② 2% peroxyacetic acid solution;
③ 0.1 mol/L NaOH solution;
③ 0.1 mol/L NaOH solution. ④ Sterile water.

The basic procedure of aseptic hysterectomy can be divided into the following steps:

A. Dislocate and execute the pregnant rats, and sterilize the whole body by soaking in iodophor for 5-10 seconds;

B. With the pregnant rat's abdomen facing upwards, the lower abdomen is cut open with surgical scissors to expose and separate the uterus bilaterally;

C. Clamp the cervix and fallopian tubes with 3 hemostats, cut the cervix along the hemostats at the vaginal end of the uterus and the fallopian tubes along the hemostats at the fallopian tube end;

D. Place the retractor cord over the hemostat handle, place the hemostat and uterus into a germicidal trap filled with 2% peroxyacetic acid disinfectant, close the outer cap of the germicidal trap, and open the inner cap of the germicidal trap after 20 seconds inside the isolator, gently pull the retractor cord to retract the hemostat, and allow the uterus to pass through the trap into the isolator;

E. Place the uterus in 0.1 mol/L NaOH solution to neutralize the peroxyacetic acid and then in sterile water to wash the lye;

F. Cut open the uterus, remove the fetal rat, and wipe the nose, mouth, and whole body amniotic fluid of the suckling rat with sterile gauze. When the whole body of the rat turns red, it means that the blood in the placenta has completely flowed into the rat's body, and the rat is able to breathe on its own, then cut off the band of the rat at a distance of 1.5 cm from the abdomen, and put the rat into a rat box lined with gauze;

G. Connect the surgical isolator with the feeding isolator and transfer the mice into the feeding isolator.
