The ELISA method can be used to detect human IL-15 in culture supernatants and serum samples. This method can also be used for the detection of monkey IL-15, but the data must be corrected using recombinant monkey IL-15 as a standard Author: J.E. Colligan et al.

basic program Material 包被抗体：纯化的抗人IL-15单克隆抗体Mill (Genzyme) V P B S PBS/Tw een 20: 0 . 0 5 % (V W ) Tween 20, PBS 配制 样品稀释液：含 有 0.5 mol/L N a C l和 （对 血 清 样 品 ） 5 % (V A O 普通鼠血清 (Life Technologies) 的 PBS/Tween > 2 0 jug/ml 重组人 IL-15 标 准 品 （Genzyme) 待测培养上清或血清样品（一70°C 保存以维持稳定性） 二抗 ：兔 抗 人 IL-15多 克 隆 抗 体 （如 Immmiex) 二抗稀释液：含 有 （对血清样品） 5 % (V /V ) 普 通 小 鼠 血 清 （L ifeT ech n ologies) 的 PBS/Tween 辣根 过 氧 化 物 酶 （H R P ) 偶联的驴抗兔IgG (JacksonImmunoresearch) T M B 过氧化物酶底物溶液（Kirkegaard& Perry) lm ol/L H 3P〇 4 (室温保存不超过1 年） 检测板： 9 6 孔 平 底 微 量 板 （如 Nunc MaxiSorp) 微 量 板 洗 漆 仪 （如 Nunc; 可选） 微量滴定板读板仪（酶联仪） ELISA 分 析 软 件 （如 BioMetallics 的 DeltaSoft) 1 . 用 P B S 稀释包被抗体至浓度为5jug/ml (使用新鲜稀释的包被抗体）。加 入 100/^1到 检测板每个孔中，盖上盖子， 2〜8°C 过夜孵育。 2 . 倒出板中液体，在微量板洗涤仪上用室温的P B S /T w een 洗 板 6〜8 次 ，每次洗涤要 确保孔中加满洗涤液。甩出残留液体。也 可 以 用 洗 瓶 加 P B S /T w een 到孔中并甩出 液体。 3 . 在包被好的微孔板上加入IOOfxl样品 。将 IL-15标 准 品 稀 释 到 800pg/m l ，在第一排 第一孔加入10(^1 (仅使用新稀释的标准品）。轻轻吹打混勻。用新的吸头移入100/xl 到同一列的下一孔中。按 照 1 : 2 倍比稀释重复7 孔 ，第 8 孔作为稀释液对照。 4 . 每个待测样品，第一个孔中加入100^1，然后进行4 次倍比 系 列 稀 释 （每孔最终体积 应 为 100M1)。对于血清样品，每个样品稀释液中应含有5 % 普通小鼠血清。 血 清 样 品 可 能 含 有 能 识 别 包 被 抗 体 的 抗 鼠 I g 抗 体 。 5 . 盖上盖子，室温孵育60min。按 步 骤 2 洗板。 6 . 用抗体稀释液按1 : 2 0 0 0 稀释二抗，每 孔 加 100^1。血清样品应含有5 % 普通小鼠血 清 。盖上盖子，室温孵育60miii。按 步 骤 2 洗板。 7 . 用 PBS/T w e e n 按 1 : 1 0 0 0 稀 释 H R P 偶联的驴抗兔IgG，每 孔 加 100M1。 盖上盖子， 室温孵育60min。按 步 骤 2 洗板。 8 . 根据生产商说明准备好T M B 过氧化物酶底物溶液，每孔加IOO1 LtU室温孵育lOmin。 每 孔 加 IOOiUd l m 〇l/L H 3P O 4终止显色过程。 9 . 用酶联仪在O D 45。处读数并用DeltaSoft软件或相似软件分析数据。也可以手绘标准曲

The concentration of IL-1 5 in the samples to be measured can be determined from the standard curve by drawing a standard curve with a line or with simple graphic software.
