Operating on an open workbench

Summary

Clean and wipe the work area, place the reagent bottles, pipette, etc., on the bench (Figure 6.5), and make preparations.

If necessary. If necessary, flame sterilize the items to be used. Keep the work surface clean. Finally. Organize the work surface. Wipe the work surface with 70% ethyl alcohol.

Pipette or ear bulb (Figures 5.5 and 6.6); 70% ethyl spray bottle, lint-free cotton swabs; absorbent paper towels; pipette tubes with water and disinfectant (see Section 7.8.5); scissors, ethanol-resistant markers, notebooks, pens, lab instructions, etc.

Procedure.

1. Scrub the work surface with 70% ethanol.

2. Thaw the culture medium from the cold box or freezer and scrub the bottle with ethanol. Put the items used in the bottle on the table and put the other items on the side.

3. Select pipettes and place them on the side of the bench where they are easily accessible.
. (a) For Borealis straws, open the box and place the lid on top or to the side. The opening is downwards.
(b) If it is a plastic straw, remove the outer packaging, according to the specifications, type classification, and put the wrapped straws on the shelf or in the box.

4. Select other glassware, plastic products, instruments, etc. that you need. Put them close by.

5. Sear the necks of the bottles by quickly rotating them in the flame and then loosening the caps.

6. Select the pipette.
. (a) In the case of Borealis straws. When removing straws from the box, keep the straw to be removed parallel to the other straws in the box and touch them as little as possible. In particular, the head of the pipette should not be used if it touches the ends of the other pipettes still in the box. Insert the zinc pipette into the pipette with the straw folded to your outside. Fold the pipette to your outer side, over the upper part of the scale. In this way, the part of the pipette that goes into the test or culture flask will not be contaminated (Figure 6.9).
(b,) In the case of plastic straws, open the outside of the package and fold it outwards. Then peel down. Nearly to the F side, insert the end of the suction bone into the ear bulb or pipette and remove the straw from the wrapper. Do not allow the pipette to touch the outside of the package or a non-sterile work surface, and finally, dispose of the wrapper in the waste basket.

7. Move the straw back and forth along the flame from bottom to top, making 180' rotations as you go, this process should only last 2-3 s. Otherwise, the straw will be too hot. Otherwise, the straw will be too hot. The purpose is not to sterilize the straw, but to hold the dust that falls on the straw. If the straw touches something or contaminates the straw in some way, it should be disposed of in the sterilizer. Do not attempt to re-sterilize the straw by burning it.

8. Insert the pipette into the ear bulb or pipette with the pipette pointing outward from the operator and the hand on the top of the scale so that the part of the pipette that goes into the reagent bottle or culture flask does not become contaminated.

Safety Tips: Don't use too much force to insert the straw into the earball, otherwise it will break the straw, don't use too much force to insert the straw into the earball, otherwise it will break the straw. Do not burn the plastic straws.

9. When loading pipettes into the lugs or pipettes, keep the pipettes at an appropriate angle and be careful not to let the tips of the pipettes touch the outside of the reagent bottle or the test tube box. Paying attention to the position of the pipette is not easy for beginners. However, it is a necessary condition for success and can be achieved in practice.

10. With the tip of the pipette pointing away from the operator, remove the cap of the first reagent bottle. Hold it in the curve formed between the little finger and the palm of the hand (Fig. 6.6) and, if several bottles are to be pipetted, place the bottles in an upright position. Tilting the bottles prevents the hand from moving over the open tops of the bottles. If it is difficult to pipette with the powder in your hand. {Place the lids on the bench with the lids facing downwards. If the bottles need to be kept constantly open, they should be placed as flat as possible on the bench or in a rack.

11. Cauterize the neck of the bottle.
Note: If Duran bottles are to be cauterized, do not use a blowtorch. Do not use a blowtorch.

12. Tilt the bottle toward the pipette to prevent the handle from moving onto the open bottle.

13. Suck out the desired amount of liquid and keep it from flowing.

.4. Cauterize the flask by tipping it and recapping it.

15. Remove the cap from the bottle receiving the liquid, cauterize the neck of the bottle, add the liquid - re-cauterize the neck of the bottle - and put the cap back on.

16. When the operation is completed, tighten the cap.

17. When the experiment is complete, remove the mother liquor from the bench, leaving only the bottles that are still needed.
