Mycoplasma Staining Solution (DNA Fluorescent Staining Method) is a reagent for in‑situ staining to detect mycoplasma or other prokaryotes in cultured cells. It is mainly used to verify mycoplasma contamination in cultured cells.
Bacterial, yeast and mold contamination in cultured cells can be observed under an optical microscope. However, mycoplasma contamination cannot be visualized under an optical microscope and must be examined by dedicated methods. Common mycoplasma detection approaches include mycoplasma isolation and culture, mycoplasma‑specific enzyme assay, RT‑PCR assay, and DNA fluorescent staining. This product detects mycoplasma via DNA fluorescent staining, enabling rapid, effective and highly‑sensitive identification of mycoplasma contamination.
Operating Procedures:
1. Add Mycoplasma Detection Staining Solution at a ratio of 2 drops per mL of culture medium.
2. Incubate at room temperature for 15‑30 minutes.
3. Observe under a fluorescence microscope (wavelength: 360~400 nm).

Figure 1 shows the condition without mycoplasma contamination. Figure 2 shows mild mycoplasma contamination; particulate mycoplasma particles are indicated by arrows. Figure 3 shows severe mycoplasma contamination, where abundant particulate mycoplasma can be observed.

2. Fluorescent dyes are susceptible to photobleaching. It is recommended to complete detection on the same day after staining.
