Visão geral

V5 Tag Peptide is a commonly used to elute V5-tagged fusion proteins from Anti-V5 Tag antibodies coupled with magnetic beads or other resins via competitive bindings during immunoprecipitation. This product elutes the V5-tagged proteins specifically and the eluted sample does not contain Anti-V5 Tag antibodies, which effectively eliminates their interference with the subsequent analysis such as Western Blot.V5 Tag Peptide is a synthetic peptide with 14 amino acids (GKPIPNPLLGLDST).The main characteristics of V5 Tag Peptide are listed below

Precautions:

The stock solution of this product should be aliquoted and stored at -20℃ or lower temperature to avoid repeated freeze-thaws.This product is for R&D only. Not for drug, household, or other uses. For your safety and health, please wear a lab coat and disposable gloves during the operation.

Instructions for Use:

1.only trace amounts of peptides are deposited in the tube during the lyophilization. Before opening the tube, we recommend centrifuging the tube at approximately 8,000-12,000×g for 10-30 seconds to collect the peptides to the bottom of the tube.2.For P9813-25mg, dissolve the peptide in TBS completely by pipetting or flicking the tube gently. Do not vortex to avoid the denaturation of the peptide.3.Competitive elution of V5-tagged proteins from immunoprecipitated samples: This is a non-denaturing method with high elution efficiency and the eluted sample does not contain the Anti-V5 Tag antibody.a.Preparation of the V5 Tag elution buffer: Based on the experimental needs, prepare an appropriate amount of elution buffer by diluting the V5 Tag Peptide stock solution to 150μg/ml with TBS.b.Add 100μl of V5 Tag elution buffer (150μg/ml) to each immunoprecipitated sample and incubate on ice or at 4℃ with shaking for 0.5-2h. To increase the elution efficiency, the incubation time can be extended or the elution can be repeated. The volume of V5 Tag elution buffer is generally 5 times of the initial volume of the gel suspension or magnetic bead suspension.c.Centrifuge at 6,000×g for 30 seconds at 4℃ and carefully transfer the supernatant to a new tube. The supernatant contains the V5-tagged proteins or their protein complexes.4.Store the eluted V5-tagged proteins or protein complexes at 4℃ for immediate use, or -20℃ for long term storage.

Application Protocols

No item-specific, tested application protocols are provided for this SKU.

General literature practices for V5 epitope peptides (for user adaptation only):

  • Antibody competition control:
    • Prepare peptide at 0.1–1 mg/mL in PBS. Pre-incubate the primary anti-V5 antibody with 10–100 µg/mL peptide for 30–60 min at room temperature, then proceed with Western blot or immunostaining. Optimize concentrations based on antibody affinity.
  • ELISA calibration:
    • Immobilize anti-V5 capture on plates, block, then titrate peptide from low nM to µM to assess dynamic range and specificity; detect with secondary recognizing the capture antibody Fc.
  • SPR/BLI kinetics:
    • Load anti-V5 onto sensor, inject peptide at graded concentrations in running buffer with low detergent (e.g., 0.005–0.01% Tween-20) to determine association/dissociation parameters.

These are general suggestions and may not fit all use cases. Follow your internal SOPs and validate conditions with small-scale pilots. For definitive, product-specific protocols, consult the CoA or contact technical support.

Biological Roles

This product is an epitope peptide for laboratory use and does not have an intrinsic biological role in living systems when used as a synthetic reagent.

General background (literature):

  • The V5 epitope is derived from a viral protein sequence and is widely adopted as a heterologous tag to facilitate detection and purification of engineered proteins using anti-V5 antibodies.
  • As a free peptide, it can mimic the epitope recognized by anti-V5 antibodies, enabling specificity controls and calibration.
  • The peptide itself is not intended to modulate biological pathways; any observed biological effects typically arise from experimental context (e.g., high local concentration, charge interactions) rather than from a defined physiological function.

Practical considerations:

  • In cell-based assays, avoid exposing live cells to high concentrations of peptide unless explicitly required by the experimental design, as non-specific uptake or osmotic effects can occur (general consideration).
  • For biochemical assays, the peptide is useful as a defined analyte to benchmark binding kinetics in surface plasmon resonance or biolayer interferometry when paired with anti-V5 capture surfaces.

Item-specific biological origin, sequence, or modifications are not specified; consult CoA if such details are critical for your application.

Buffer Applications

This item is a peptide reagent, not a buffering agent, so it is not typically used to establish or maintain pH.

Practical notes (general, literature):

  • Reconstitution is commonly performed in non-buffered water or mild buffers (e.g., PBS, HEPES) selected for the downstream assay. Buffer choice should preserve antibody–epitope interactions and maintain peptide stability.
  • Typical working solutions are prepared at 0.1–1 mg/mL, then diluted as needed for competition or calibration experiments. The optimal concentration depends on assay affinity and is not specified for this item.
  • Avoid extreme pH (<5 or >9) unless necessary for solubility studies, as peptides may undergo deamidation or backbone hydrolysis over time under harsh conditions.

If you require a specific buffer recipe, pH range, or ionic strength for this peptide, consult your assay SOP and the product CoA; item-specific buffer recommendations are not provided.

Green Alternatives

Green chemistry considerations for epitope tag workflows (general):

  • Choice of tag vs. protein fusions:
    • Peptide tags are short and typically have lower resource and energy footprints in synthesis and purification compared with large protein affinity tags produced in cells.
  • Solvent minimization:
    • Favor aqueous buffers for dissolution and handling; avoid high percentages of organic solvents during reconstitution to reduce VOC emissions and hazardous waste.
  • Reagent economy:
    • Use micro-scale antibody validation with peptide competition to reduce consumption of antibodies and sample materials.
  • Solid-phase synthesis waste:
    • Peptides are commonly made by SPPS which uses DMF/DCM and coupling reagents. When selecting suppliers or custom lots, inquire about greener SPPS practices (e.g., NBP/PC or EtOAc solvent platforms, recyclable resins, optimized coupling to reduce excess reagents).

Comparison (general):

  • Peptide competition controls vs. plasmid-based expression controls:
    • Peptide controls avoid cell culture, antibiotics, and energy-intensive incubations, reducing biohazard waste.
    • Tradeoff: SPPS-derived peptides carry embedded synthesis impacts; using concentrated stocks and aliquoting minimizes repeat syntheses and shipments.

For this item, no specific green certifications or production process details are provided; consult the supplier if sustainability metrics are required for your project.

Pharmaceutical Uses

For research use only; not for human or veterinary use, and not for diagnostic or therapeutic procedures.

Context in pharmaceutical R&D (general):

  • Analytical reagent: Epitope peptides are used as reference materials or system suitability standards in immunoassays that detect V5-tagged research proteins during discovery and process development.
  • Process characterization: Can serve as a specificity control to verify that anti-V5 capture or detection steps do not cross-react with host-cell proteins in analytical workflows.
  • Conjugation studies: Employed in method development for peptide–antibody interaction characterization, without being part of a drug product.

Regulatory note:

  • No pharmacopeial monograph is associated with V5 Tag Peptide. Any GMP-related use would require project-specific qualification, impurity profiling, and change control. Such qualifications and grades are not specified for this item.
Physical Properties
  • Appearance: Not specified for this item; refer to CoA/Spec Sheet. (Peptide tags are commonly supplied as lyophilized powders or films.)
  • Molecular weight, formula, and exact composition: Not specified for this item; refer to CoA/Spec Sheet.
  • Melting point/decomposition: Not specified for this item. Peptides typically do not have a sharp melting point and may decompose on heating (general literature).
  • Solubility (general, literature guidance):
    • Short hydrophilic peptides such as epitope tags are often soluble in aqueous buffers (water, PBS) at mg/mL levels. Solubility can vary with sequence, counterion (e.g., TFA), pH, and ionic strength.
    • If initial dissolution is slow, brief sonication or adjusting pH/ionic strength can help. For highly hydrophobic sequences, a small percentage of organic co-solvent (e.g., acetonitrile) may assist, followed by dilution into buffer (general practice).
  • pI/logP/pKa: Not determinable without the exact sequence; Not specified for this item.
  • Refractive index/density: Not applicable for solids; Not specified for this item.
  • Hygroscopicity (general): Many peptides are mildly hygroscopic; minimize ambient exposure to maintain stability and accurate massing.

Important: For any quantitative property needed for your method validation (e.g., exact MW for mass spectrometry), consult the item-specific CoA or request the analytical report.

Quality and Grades
  • Grade/Purity: Not specified for this item; refer to CoA/Spec Sheet.
  • Research Use Note: For research use only.

What quality typically means for synthetic peptides (general, literature):

  • Purity is often reported as HPLC area percent and verified by mass spectrometry. Common research grades include crude, desalted, and >90–98% HPLC-purified. Absence of an item-specific purity means end users should check the CoA to confirm suitability for sensitive assays (e.g., quantitative ELISA controls).
  • Counterions and salt form (e.g., TFA, acetate) can impact solubility and bioassay performance; item-specific counterion is not specified.
  • Water/volatile content and residual solvents are usually characterized for QC but are not specified for this item.
  • Sequence confirmation is typically provided by ESI/MS or MALDI/MS; for this item, verification details are not specified.

Implications for use:

  • For antibody validation or calibration curves, ensure the supplied purity/identity meets your acceptance criteria. If your application requires specific termini (e.g., amidated C-terminus to mimic fusion-tag context), confirm those features on the CoA before use.
Reaction and Applications

Applications (general, literature):

  • Epitope standard: Commonly used as a positive control peptide for validating anti-V5 antibodies in Western blot, ELISA, immunocytochemistry, and immunoprecipitation workflows.
  • Competitive inhibition studies: Can be used to verify binding specificity by pre-incubating anti-V5 antibodies with free peptide.
  • Assay development: Useful for calibrating capture assays targeting the V5 epitope and for mapping antibody affinity.
  • Conjugation/immobilization: Peptides may be coupled to activated surfaces (e.g., NHS-ester resins) to create affinity matrices for anti-V5 reagent characterization.

Chemical reactivity and practical notes (general):

  • Amine coupling: Free N-terminal and Lys epsilon-amines (if present) react with NHS esters under mild basic conditions (pH 7.5–8.5). If terminal modifications are present, amine availability may differ; item specifics not provided.
  • Carboxyl activation: C-terminus can be activated (e.g., EDC/NHS) for coupling to primary amines on surfaces or carriers at pH 5–6.
  • Sequence-dependent behavior: Net charge and hydropathy influence adsorption to plastics and recovery; low-binding tubes and the presence of carrier proteins (e.g., BSA in assay buffers) can reduce loss.
  • Analytical verification: HPLC and LC–MS are standard to confirm integrity after storage or conjugation.

Note: Manufacturer applications are not listed for this SKU; the above use-cases reflect general literature practices for V5 epitope peptides.

Solvent Selection

This product is a peptide tag and not a general-purpose solvent. Solvent selection pertains to its reconstitution/dilution.

General guidance (literature):

  • Primary solvents: sterile water, PBS, or other physiological buffers (pH 6.5–7.5) are typically suitable for short epitope peptides. Adjust pH if sequence contains multiple basic or acidic residues.
  • Co-solvents: For difficult dissolution, use minimal volumes of acetonitrile or ethanol as a wetting agent, then dilute with buffer. DMSO can be used for stock solutions when compatible with the downstream assay; keep final DMSO ≤1–2% in bioassays.
  • Additives: 0.1% TFA or 0.1% acetic acid may improve solubility for basic peptides; low concentrations of NaCl can reduce electrostatic aggregation.
  • Filtration: 0.22 µm sterile filtration after dissolution can improve clarity and sterility if compatible with intended use.

Selection tips:

  • Choose pure aqueous buffers for antibody binding assays to avoid organic solvent effects on epitope–antibody interactions.
  • For long-term stocks, consider adding 10–50% glycerol for freeze–thaw protection if the peptide remains soluble; verify compatibility with your assay.

Item-specific solvent recommendations are not specified; consult the CoA and your SOPs.

Storage and Reconstitution
  • Storage conditions (item-specific): Store at -20°C.
  • Shipping (item-specific): Shipped in an ice chest with ice pads.

General reconstitution guidance for peptides (literature; verify against your SOPs):

  • Allow the vial to equilibrate to room temperature in a desiccated environment before opening to prevent condensation.
  • Reconstitute in sterile water or buffer compatible with your assay (e.g., PBS, pH ~7.4). Start with a small volume to achieve a concentrated stock (e.g., 1–5 mg/mL), then dilute as required.
  • If dissolution is incomplete, gentle vortexing and brief sonication can help. Adjust pH or add minimal organic co-solvent if necessary, then dilute back into buffer.
  • Aliquot immediately after reconstitution to avoid repeated freeze–thaw cycles. Store aliquots at -20°C or -80°C for extended stability. Protect from light and moisture.
  • Avoid prolonged exposure to ambient temperatures. For working solutions kept at 2–8°C, use within a few days unless stability is verified.

Item-specific details such as exact sequence, counterion, purity, and recommended solvent are not specified for this product; refer to the CoA/Spec Sheet for definitive handling instructions.
