1/2 MS medium is a widely used modified basal medium for plant tissue culture. Based on the standard MS medium formula, the concentration of macroelement inorganic salts is halved, while core organic and micro-nutrient components including trace elements, iron salts, vitamins and glycine from the original formula are fully retained. Sucrose and agar are added in matched proportions to provide a mild and balanced nutritional environment for the growth of in vitro plant cultures. It is extensively applied in sterile seedling cultivation, rooting culture of tissue-cultured seedlings, seedling hardening and rejuvenation, in vitro culture of stem segments and buds, protoplast culture, anther tissue culture and other procedures, serving as the mainstream dedicated medium for plant rooting stages.

Usage Instructions:
1. Accurately weigh 39.45 g of this product and add it to 1 L of distilled water. Stir until the powder is completely dissolved to prepare a homogeneous medium solution.
2. Sterilize via high-pressure steam at 116 °C for 30 minutes. After sterilization, place the medium in a clean environment for natural cooling before use in plant tissue culture experiments.
Precautions:
1. Store at room temperature away from light; shelf life is 3 years.
2. This plant tissue culture medium adopts a full inorganic salt formula. A small amount of precipitate may form after sterilization. Dissolve the powder in room-temperature water prior to autoclaving; hot-water dissolution is not recommended.
