5'-Nucleotidase (5'-NT) catalyzes the hydrolysis of nucleoside-5'-monophosphate to nucleoside and inorganic phosphate (Pi). Under acidic conditions, the released inorganic phosphate reacts with ammonium molybdate to form ammonium phosphomolybdate, which is subsequently reduced to blue molybdenum blue. The resulting product exhibits a characteristic absorbance maximum at 660 nm. The activity of 5'-nucleotidase can be quantified by monitoring the change in absorbance at this wavelength.
