ABTS is oxidized by oxidizing agents to form a stable blue-green cation radical (ABTS⁺·), which exhibits characteristic absorbance maxima at 405 nm or 734 nm. In the presence of antioxidants, the generation of ABTS radicals is inhibited, resulting in decolorization of the reaction system and a consequent decrease in absorbance at 405 nm. Within a certain range, the change in absorbance is proportional to the extent of radical scavenging. Thus, the ABTS radical scavenging capacity of a sample can be characterized by the decrease in absorbance. Trolox is provided as a positive control to quantify the scavenging capacity of antioxidant substances.
