Intended Use
For acid - fast fluorescence staining of mycobacteria and other bacteria.
Test Principle
This product utilizes the characteristic that mycobacteria can bind to AIE fluorescent dyes and are not easily washed off by acidic decolorizing solution. As a result, mycobacteria show significant fluorescence under the observation of a specific channel of a fluorescence microscope, while non - mycobacterial groups show no or very weak fluorescence, thus distinguishing and detecting mycobacteria. This staining solution can be used to observe mycobacteria under a low - power microscope, and their morphological structure is clearly distinguishable.

Please read this instruction manual carefully before using this product.
1.Smear the sample to be tested on a glass slide, heat and fix it to form a coating film of appropriate thickness.
2.Add AIE staining solution until the coating film is covered, place it on a 60℃ heating plate and stain for 3min (additional staining solution can be added if necessary to prevent the staining solution from drying), then rinse with distilled water.
3.Add acidic decolorizing solution until the coating film is covered, decolorize for 3min, rinse with distilled water, and cover with a cover slip.
4.Observe the smear through channel B of the fluorescence microscope (Excitation: 460 - 490nm; Emission: 510 - 550nm). Mycobacteria show bright green and their morphological structure is clearly distinguishable, while non - mycobacterial groups show no color or very weak green.
Precautions
1.Cap the reagent bottle after taking the reagent to prevent reagent volatilization and contamination.
2.When using this product, wear disposable gloves to avoid contact with skin and eyes. Once in contact, immediately rinse with a large amount of water. If swallowed or splashed into the eyes, please seek medical attention in time.
3.All samples, biological reagents and experimental materials should be regarded as potentially infectious and must be disposed of as medical waste in accordance with local regulations.
