Alcian Blue (also known as Alcian blue) is a copper phthalocyanine conjugated dye originally used for textile fiber dyeing. This cationic dye binds to acidic groups, i.e., Alcian blue forms insoluble complexes with anionic groups in tissues such as carboxyl and sulfate groups. Alcian blue consists of a central copper‑containing phthalocyanine ring linked via thioether bonds to four isothiouronium groups. These isothiouronium groups are moderately alkaline, giving Alcian blue its cationic character. At pH 2.5, carboxyl groups in tissues ionize, carrying a negative charge, and form salt bonds with the cations in Alcian blue, thereby staining acidic mucus substances containing carboxyl groups (sulfated mucins and sialomucins).
Alcian Blue Staining Solution (pH 1.0) utilizes different pH levels of the dye to distinguish classes of mucus substances. At pH 1.0, carboxyl groups (COOH) do not stain, while sulfate groups (OSO₃H) stain. At pH 2.5, carboxyl groups stain well but sulfated mucins stain poorly. At pH 1.0, sulfate groups in tissues ionize, carrying a negative charge, and form salt bonds with the cations in Alcian blue, staining tissues containing sulfate groups (e.g., sulfated mucus substances). Neutral mucins (such as neutral mucins in gastric mucosa and Brunner’s glands) do not react with Alcian blue.

Materials to Be Prepared by User
Graded ethanol series, distilled water
Instructions for Use
Deparaffinize in xylene, rehydrate through a graded ethanol series, and rinse in distilled water.
Immerse in Alcian Blue Acidification Solution for 5 minutes.
Stain in Alcian Blue Staining Solution for 30 minutes. Rinse under running water.
Rinse briefly with Alcian Blue Acidification Solution.
Without water rinse, blot off excess Alcian Blue Acidification Solution using filter paper.
Counterstain in Nuclear Fast Red Staining Solution. Rinse under running water.
Dehydrate routinely through ethanol, clear in xylene, and mount with neutral balsam.
Precautions
Use 10% neutral formalin as fixative.
This method can distinguish sulfated mucins from proteoglycans; its pH = 1.0.
Opened reagents should be used within 6 months after opening. Tighten the cap immediately after each use to prevent evaporation or deterioration.
