Descripción general

Amido Black 10B can be used to detect proteins on blotting membranes. When the amount of transferred protein exceeds 50 ng per band, dark blue bands appear against a light blue background. For the determination of internal protein sequences, Amido Black is the most commonly used stain for protein sequencing and in situ protein cleavage. 
Amido Black 10B Staining Solution (0.1%) is mainly composed of Amido Black 10B, weak acid and deionized water. It is suitable for PVDF membranes, nitrocellulose membranes and nylon membranes. It is characterized by mild staining conditions, which minimize protein elution. This product is for research use only and shall not be used for clinical diagnosis or other non-research purposes. 
Materials Required
  • Distilled or deionized water, 5% acetic acid
  • Shaker 
Protocol (For Reference Only)
  1. Place the blotting membrane in a suitable container and wash with distilled water 3 times, 5 min each.
  2. Immerse in Amido Black 10B Staining Solution (0.1%) for 1 min or longer.
  3. Destain in 5% acetic acid twice on a shaker at room temperature, 1 min each.
  4. Rinse 2–3 times with distilled water, PBS or other appropriate solution, 10 min each.
  5. Document or photograph the results appropriately.
