β-Galactosidase (β-GAL, EC 3.2.1.23) is a hydrolytic enzyme widely present in animals, plants, microorganisms, and cultured cells. It catalyzes the hydrolysis of β-galactosidic bonds in β-galactoside compounds and additionally possesses transgalactosylation activity. β-GAL not only releases stored energy for rapid plant growth but is also involved in normal polysaccharide metabolism, cell wall component turnover, and the hydrolysis of terminal galactose residues in polysaccharides, glycoproteins, and galactolipids during cell wall degradation in senescence, thereby releasing free galactose. β-GAL catalyzes the hydrolysis of p-nitrophenyl-β-D-galactopyranoside to generate p-nitrophenol, which exhibits a maximum absorption peak at 400 nm. The activity of β-GAL is calculated by measuring the rate of increase in absorbance at this wavelength.
