Descripción general
Bacterial Freezing Medium can be used for cryopreservation of various common bacteria.Generally, strains can be stored for 12 months at -20 °C, and for long‑term storage at -70 °C or -80 °C.This product has been autoclaved. Simply add the medium to the bacterial pellet to be preserved and mix well. This product is for research use only and is not intended for clinical diagnosis or other purposes.
Materials Required
Sterile centrifuge tubes or cryovials, pipette tips
Protocol (For Reference Only)
Culture the bacteria in liquid medium such as LB to the logarithmic growth phase.
Collect the bacterial pellet by low‑speed centrifugation using a sterile centrifuge tube or cryovial, and discard the supernatant.
Add an appropriate volume of this product to resuspend the bacteria, typically 0.2–1 mL of Bacterial Freezing Medium, and mix thoroughly.
Store the bacteria at -20 °C or -80 °C.
For recovery of frozen bacteria, scrape solid ice crystals from the surface of the frozen stock using a sterile loop, sterile wooden stick (toothpick), or pipette, and streak onto a suitable agar plate (e.g., LB agar plate).Handle carefully and avoid thawing the frozen stock.
Precautions
Bacterial Freezing Medium must be used in a laminar flow hood to avoid microbial contamination.
The bacteria must be thoroughly mixed with Bacterial Freezing Medium; otherwise, it cannot protect the bacteria during cryopreservation.
Fresh bacteria in logarithmic phase give the best freezing results, but non‑logarithmic or non‑freshly cultured bacteria can also be preserved with this medium.
Do not inoculate after directly thawing the frozen stock.
For your safety and health, please wear a lab coat and disposable gloves during operation.
Please use the reagent promptly after opening to avoid affecting experimental performance.
