Descripción general

Chondroitin Sulfate (CS) is a type of sulfated glycosaminoglycan. Its carbohydrate chain consists of alternating disaccharide units of glucuronic acid and N-acetylgalactosamine. It exists in the extracellular matrix of connective tissues, such as bone, cartilage, skin, ligaments, and tendons, and exhibits excellent biocompatibility and biodegradability. CS demonstrates antioxidant, antithrombotic, anti-inflammatory, and immunomodulatory effects, and can be used in the prevention and treatment of osteoarthritis, skin wounds, central nervous system diseases, cancer, cardiovascular and cerebrovascular diseases, and ophthalmic diseases.
CSMA is a photosensitive polymer material obtained by modifying chondroitin sulfate (CS) with methacrylic anhydride (MA). It can be used in combination with blue light or UV photoinitiators and undergoes cross-linking and curing under blue light or UV irradiation. This polymer possesses excellent biocompatibility, biodegradability, antioxidant, antithrombotic, anti-inflammatory, and immunomodulatory properties, with simple curing conditions. It can be applied in fields such as bioprinting, 2D/3D cell culture, tissue engineering and organ construction, cartilage regeneration, and wound dressings.

Instructions for Use

1. CSMA can be used in combination with o-nitrobenzyl alcohol-modified polymers such as PEGNB, HANB, and GelNB. Polymerization and cross-linking are initiated by photoinitiators LAP or NAP under light irradiation, and high-strength hydrogel materials can be obtained within seconds.

2. CSMA can be used in combination with methacryl-modified polymers (e.g., CMCMA, GelMA, HAMA, ChMA) and allyl-modified polymers (e.g., F127DA). Polymerization is initiated by photoinitiators under light irradiation to fabricate hydrogel materials with diverse physicochemical properties.

3. CSMA can be used alone, with polymerization and cross-linking initiated by photoinitiators under light irradiation.

Preparation Method

1. Preparation of photoinitiator stock solution: Dissolve an appropriate amount of NAP or LAP to prepare a solution with a concentration ranging from 0.1 wt% to 0.25 wt%. Store at 2-8 °C away from light.

2. Weigh the required amount of CSMA into a centrifuge tube, then add the required volume of the initiator stock solution into the tube.

3. Stir the sample in the dark at room temperature or shake on an orbital shaker until completely dissolved. If the sample clumps in the solution, crush it with a spatula first; if dissolution is slow, heat and stir at 50-60 °C to assist dissolution.

Precautions

1. The higher the concentration of the prepared CSMA solution, the greater the modulus of the cured gel and the shorter the curing time.

2. The recommended working concentration of CSMA shall not exceed 20 wt%.

3. The higher the concentration of the photoinitiator stock solution, the faster the curing speed of the CSMA solution, and the more brittle the cured gel.

Sterilization Methods

1. Filter sterilization (recommended): Sterilize the solution using a 0.22 μm sterile syringe filter. This method is only applicable to low-viscosity solutions. For high-viscosity solutions, pasteurization or moist heat sterilization is recommended.

2. Pasteurization: Heat the solution to 80 °C and hold for 30 min, then quickly cool to room temperature in an ice-water bath. Repeat the cycle three times.

3. Moist heat sterilization: Sterilize the solution in an autoclave at 121 °C for 8 min. After manual pressure release, quickly cool the solution to room temperature in an ice-water bath.

Note: High temperatures may degrade gel performance; strictly follow the above procedure when using this sterilization method.
