Citrate Synthase (CS) (EC 2.3.3.1) is widely present in the mitochondrial matrix of animals, plants, microorganisms, and cultured cells. As the first rate-limiting enzyme of the tricarboxylic acid (TCA) cycle, it serves as one of the major regulatory sites in this central metabolic pathway. The detection principle of this kit is as follows: CS catalyzes the condensation of acetyl-CoA and oxaloacetate to form citryl-CoA, which is subsequently hydrolyzed to produce citrate. This reaction drives the conversion of the colorless compound DTNB into the yellow-colored product TNB. TNB exhibits a characteristic absorbance at 412 nm. By measuring the rate of increase in absorbance at 412 nm, the activity of citrate synthase can be calculated.
