Human collagen type IV is a native triple helix and heterotrimeric protein containing two α1-like and one α2-like genes. Type 4 collagen forms a sheet-like extracellular matrix in multicellular tissues that exists around certain cell types like adipocytes, skeletal muscle cells, heart muscle cells, and smooth muscle cells. Human Collagen IV is extracted from washed dissected tissue into dilute acetic acid after mild pepsin treatment. The Collagen type 4 was purified by using differential salt precipitation. Purity and retention of the native helical structure were controlled by SDS-PAGE and by reaction with anti-collagen type-specific monoclonal antibodies.
