India ink capsule staining is highly effective for the observation of Cryptococcus neoformans. India ink is a premium-grade ink. A mucopolysaccharide capsule exists around the cell body of Cryptococcus neoformans, and the presence of this capsule can be demonstrated by the India ink staining method. The Cryptococcus neoformans staining solution is formulated based on the India ink recipe. After staining, the capsule of Cryptococcus neoformans displaces the colloidal carbon particles in the ink, leaving the fungal cell surrounded by a distinct, colorless, and transparent halo. This reagent is intended for research use only and is not suitable for clinical diagnosis or any other purposes.
Materials to Be Prepared by User:
Centrifuge, glass slides, cover slips, microscope
Operating Procedures (For Reference Only):
1. Centrifuge the sample and spread the sediment onto a glass slide.
2. Add a drop of Cryptococcus neoformans Staining Solution onto the sample smear.
3. Place a cover slip over the smear and avoid the formation of air bubbles.
4. Press gently with your finger to mix the sample with the staining solution and flatten the smear to a thin layer.
5. Locate the capsulated microorganisms under the low-power objective lens, then switch to the high-power objective lens or oil immersion lens for detailed identification.

Precautions:
1. Opened Cryptococcus neoformans Staining Solution within its expiration date should be used up within 6 months. Seal the container tightly as much as possible after each use to prevent volatilization or deterioration.
2. Cryptococcus neoformans Staining Solution is prone to precipitation after long-term storage. If coarse particles are observed under the microscope, the solution should be filtered before use or discarded.
3. The target samples should be as fresh as possible.
4. The purity and concentration of the samples should be appropriate; it will be difficult to detect Cryptococcus neoformans under the microscope if the concentration is too low.
5. Ensure uniform distribution of the samples and the staining solution.
6. For your safety and health, wear a lab coat and disposable gloves during the operation.
