This kit utilizes D-lactate dehydrogenase (D-LDH) to catalyze the conversion of D-lactate to pyruvate, while simultaneously reducing NAD⁺ to NADH and H⁺. The generated NADH, through the electron-transferring action of 1-mPMS (1-methoxy-5-methylphenazinium methylsulfate), reduces WST-1 to a water-soluble formazan, which exhibits a characteristic absorbance peak at 450 nm. By measuring the change in absorbance at this wavelength, the D-lactate content in the sample can be quantitatively determined.
