Environment-friendly Wax-Soaking and Dewaxing Clearing Solution, also known as tissue clearing agent, dewaxing clearing agent, dewaxing clearing solution, etc., it is a colorless, odorless, low-volatility, and biodegradable organic solvent specially developed for the processing of histopathological and cytological specimens. Free of aromatic compounds, it is an ideal environmentally friendly clearing agent that can replace xylene in tissue preparation.
Paraffin is an organic compound. Based on the principle of like dissolves like, the environment-friendly clearing solution can clear tissue specimens to facilitate wax infiltration, and can also dissolve paraffin in tissues to achieve dewaxing. The reagent is non-corrosive to instruments during use. As a high-performance green and environment-friendly product, it can greatly improve the experimental working environment and protect the physical and mental health of laboratory personnel. It is mainly used for the processing of wax-soaked tissue specimens, dewaxing and clearing, as well as clearing in cytological preparation.
Protocol (for reference only):
Routine HE Staining of Tissue Sections:
1. Dewax with Environment-friendly Wax-Soaking and Dewaxing Clearing Solution (I) for 5–15 minutes
2. Dewax with Environment-friendly Wax-Soaking and Dewaxing Clearing Solution (II) for 5–15 minutes
3. Absolute ethanol for 2–3 minutes
4. 95% ethanol for 1–2 minutes
5. 80% ethanol for 1–2 minutes
6. Water rinse for 1–2 minutes
7. Hematoxylin and eosin (HE) staining (operate according to specific experiments)
8. Dehydration (operate according to specific experiments)
9. Clearing with Environment-friendly Wax-Soaking and Dewaxing Clearing Solution (I) for 2–5 minutes
10. Clearing with Environment-friendly Wax-Soaking and Dewaxing Clearing Solution (II) for 2–5 minutes
11. Mounting with mounting medium
Tissue Processing (Tissue Processor):
1. Fixation with 10% neutral buffered formalin (I) for 2 hours
2. Fixation with 10% neutral buffered formalin (II) for 2 hours
3. 80% ethanol for 45 minutes
4. 95% ethanol (I) for 45 minutes
5. 95% ethanol (II) for 45 minutes
6. Absolute ethanol (I) for 45 minutes
7. Absolute ethanol (II) for 45 minutes
8. Clearing with Environment-friendly Wax-Soaking and Dewaxing Clearing Solution (I) for 1 hour
9. Clearing with Environment-friendly Wax-Soaking and Dewaxing Clearing Solution (II) for 1 hour
10. Wax infiltration at 65℃ (I) for 1 hour
11. Wax infiltration at 65℃ (II) for 1 hour
12. Wax infiltration at 65℃ (III) for 2–3 hours
Sample Requirements: Tissues must be adequately dehydrated before use, whether for tissue wax infiltration or clearing after staining.
