Animal model of urethral hypospadias

Summary

Hypospadias is a urethral malformation in which the opening of the urethra fails to reach its normal position due to abnormal development of the anterior urethra, which is often complicated by hypospadias, striated developmental anomalies, and hernia, and it is a common congenital malformation of the male urinary system. The development of the mouse and human urethra shows strong similarities, such as the fusion of two epithelial margins, the closure of an epithelial suture midline, and the ensuing cellular remodeling, so mice are usually chosen as the experimental animal model for urethral hypospadias.

Materials and Instruments

Equipment:
① Common surgical instruments;
② stereomicroscope.
Reagents:
① Estradiol benzoate.

Move

A. Mice were paired in 2:1 pairs, and the first day of pregnancy was observed to be the first day of pregnancy. Mice with good motherhood and feeding experience were also selected as surrogate mice. The surrogate mice became pregnant 2-4 days earlier than the pregnant mice in the test group.
B. Pregnant mice were injected subcutaneously with estradiol benzoate 5 mg/(kg-d) every day from 8:30 to 9:00 p.m. for 12-16 days of gestation, and the females were delivered by C-section on 21 days of gestation after decapitation, and the surviving pups were given to the suckling mice for feeding.
C. Newborn male mice were observed under the stereo microscope for the development of the penile prepuce, the position of the urethral orifice, the urinary discharge, and the development of the nipples, and were weighed and the distance of the anogenital joints was measured. The newborn male mice were weighed and the distance between the anogenital nodes was measured.
D. Male mice were taken and dissected on the same day after birth to observe the position of the testes, the development of the prostate gland, and the delayed degeneration of the nipples.
E. Surviving mice were observed for hypospadias and cryptorchidism at 4 weeks after birth, and the malformation was determined at the same time.
