Blood plate preparation experiment

Summary

It is suitable for all kinds of bacterial growth, and should be inoculated with blood plate for the separation of general bacterial test specimens. This experiment is from Mudanjiang Medical College, undergraduate 5-year laboratory guide for testing majors.

Principle

Either sheep's blood or rabbit's blood can be added to the broth agar. The amount of blood used is 5-7%. In addition to observing the morphology of the colony on the blood plate, it can also determine the hemolysis, and the complete destruction of red blood cells in the medium around the colony is β-hemolysis. Green color around the colony is α-hemolysis. Hemolysis can be observed under microscope with low magnification.

Ingredients:

Beef extract: 1000 ml

Peptone: 10 g

Sodium chloride: 5 g

Agar: 15 g

Decontaminated and defibrinated sheep or rabbit blood: 60 ml

PH 7.2~7.4

Preparation: Mix the first four components, heat and melt, correct PH, place in a triangular flask, autoclave (121 ℃ for 15min) cooled to 50 ℃, add sterile blood, rotary fully shaken well and tilted flat. The blood agar layer was 4mm thick.
