Determination of urinary sodium experiment

Summary

Urinary sodium (Na) is the measurement of the concentration of sodium ions in the urine over a 24-hour period can be used to determine a measure of an individual's daily sodium intake.

Principle

The protein in the urine was precipitated with anhydrous ethanol to obtain protein-free urine filtrate, which was then combined with potassium pyroantimonate to produce sodium pyroantimonate precipitate. Finally compared with the standard tube to find the content of urinary sodium, the reaction formula is as follows: Nacl + K [Sb (OH) 6] → Na [Sb (OH) 6] + KCl

1. Take 0.1 of urine into a centrifuge tube, add 1.9 ml of anhydrous ethanol, shake vigorously, and centrifuge for 10 minutes to precipitate, then take the supernatant and operate as follows. 2. After the above operation, mix well and compare the color immediately:


2. After the above operation, mix well and immediately compare the color, use 721 spectrophotometer at 520 to compare the color, adjust the zero with the blank tube, read the optical density of the standard tube and the measuring tube, and calculate according to the following formula:

Sodium p.m./mlx30 minutes urine volume (ml) = total urine sodium (p.m./ml)

Note: 0.025 indicates the content in the standard solution (0.1 ml of urine diluted to 2 ml with 1.9 ml of anhydrous ethanol, the actual dilution is only 0.5 ml, so it is 0.025) 0.075 indicates the number of milligrams of sodium in the standard solution (1 ml=0.15 p.m./ml of sodium, 0.5 ml=0.075 p.m./ml of sodium).

Caveat

⒈ Add anhydrous ethanol should be vigorously shaken, so that the precipitation is formed quickly in order to make the protein precipitation particles uniform.Piece of the operation is completed immediately after the colorimetric, leaving the particles for a long time so that the results will be affected by the coarsening of the particles.
