Experiments on quantitative traits in Drosophila

Summary

Drosophila quantitative trait experiments can be applied to: (1) characterize the inheritance of quantitative traits; and (2) learn to estimate inheritance.

Principle

In organisms, any trait that is countable, measurable, measurable, etc. and can be described in numerical form is called quantitative character. Quantitative traits are mostly controlled by multiple genes. Generally, the number of genes controlling the same trait is large, while the role of each gene is small and easily influenced by the environment. The phenotypic variables of a population are usually continuously distributed.

Move

1. 20 virgin and 20 male flies were randomly selected from a cross between 18# and 22# strains of Drosophila, anesthetized with ether, and the number of setae on the fourth and fifth abdominal plates of ♀ and ♂ flies were counted under a 40x microscope, and labeled with a pinky tube (indicating the sex, and the total number of setae on the two abdominal plates).


2. Select two ♀ and two ♂ flies with the highest and lowest number of setae respectively from the above 20 flies.


3. Match 1 each of the ♀ and ♂ with the highest number of bristles into a hybrid combination, 1 each of the ♀ and ♂ with the second highest number of bristles into a hybrid combination (as a back-up), 1 each of the ♀ and ♂ with the lowest number of bristles into another combination, and 1 each of the ♀ and ♂ with the second lowest number of bristles into a hybrid combination (as a back-up).


4. Put the hybrid combination, put in the 25 ℃ incubator culture 3 - 4 days, the parents pour away.


5. After two weeks of cultivation, pour all the adults out of the test tube for anesthesia, observe the number of small setae of ♀ and ♂ flies respectively, make records and statistics.


6. Group the obtained number of small setae and determine the group spacing.

Caveat

Be patient and observant during the experiment, and be extra careful during data counting to try to avoid errors.
