Experiments on radiological safety measures

Summary

This experiment is based on the "Guide to Cellular Experiments", translated by Huang Peitang et al.

1. All 32P labeling operations are performed in an "active laboratory" equipped with all the necessary equipment for the initial processing of protein samples. This laboratory is equipped with all the equipment needed for the initial processing of protein samples, such as: thick plastic baffles for body protection, containers for collecting liquid and solid 32P waste in a plastic cover, a cell culture incubator, a bench-top low-speed centrifuge and a Geiger counter.

2. wear a lab coat and gloves throughout the experiment. The work area was checked with a Geiger counter before the experiment to make sure it was not contaminated. The table was covered with plastic-backed blotting paper to absorb occasional spills.

3. Conduct a simulation experiment with non-radioactive materials to check procedures and ensure that materials needed for labeling experiments are in a convenient place on the lab table.

4. test tubes/bottles containing high 32P activity (e.g., labeling medium) should be kept in heavy-walled plastic cases before, during, and after the experiment. A Geiger counter should be left on near the work area during the experiment, and gloves should be checked at regular intervals to make sure there is no contamination. If contaminated, they should be changed immediately to avoid further spread of contamination. Wearing 32P-contaminated gloves to hold tubes/instruments is the most common way to spread contamination. At the end of the experiment, do a thorough inspection of the work area and wash off any spills.
