Summary

This method is suitable for clinical specimens or other cultures mixed with many kinds of bacteria, which can be separated and cultured to obtain single colonies for bacterial counting and pure culture. This experiment is derived from the laboratory instruction of Mudanjiang Medical College, undergraduate 5-year testing program.

(1) Curve delineation separation method:

This method is mostly used for specimens containing little bacteria or cultures taken from pharyngeal swabs and cotton swabs, the specimens or pharyngeal swabs and cotton swabs cultures are directly coated in 1/5 of the culture medium, and then inoculated with inoculation loop or directly with pharyngeal swabs (cotton swabs) for curved continuous demarcation line inoculation.

(2) Zonal isolation method:

This method is mostly used for the bacterial isolation culture of feces and other specimens containing more bacteria. First, use the inoculation ring to take the fecal specimen and spread it in the 1/4 of the culture medium, the inoculation is made by continuous regional delineation, and every 1/4 of the delineation rotate the petri dish by 90°. The inoculating ring is flame sterilized after each 1/4 stroke, and this method dilutes the culture sufficiently by zonal scribing to obtain a larger number of individual colonies at the end.

(3) Semi-quantitative counting method:

This method is for semi-quantitative bacterial counting of urine, take 0.01 ml or 0.001 ml of unprecipitated urine on a quantitative inoculation ring, draw a line in the center of the agar dish, and on the basis of the initial drawing of the line at a right angle uniformly and continuously draw a line to spread the cultures, and then rotate the dish by 90 ° and then draw the line again, so that the inoculum is completely covered by the surface of the agar.
