Equipment: fume hood, microscope, routine laboratory instruments, blood smears.
Reagents:
① formaldehyde fixative; ② saturated Sudan black B - anhydrous ethanol

The basic process of Sudan Black B method to show lipids in cells can be divided into the following steps:
A. After air-drying of blood smear, fix it with formaldehyde vapor at 37 ℃ for 2-5 min.
B. Wash it well with tap water and then distilled water, and air-dry it.
C. Stain it into saturated Sudan Black B - anhydrous ethanol solution at room temperature for 30-90 min.
D. Wash it with 70% ethanol for 2 min, and remove the floating color.
E. Re-stain it with sandy yellow for 1 min or Giemsa re-staining. F. Rinse it in tap water for 1 min. G. Check it under the microscope. E. Re-stain with sand yellow for 1 min or Giemsa re-staining.
F. Rinse with tap water for 1 min.
G. Air dry and examine by microscope.
