Principle

Koch's I medium is a synthetic medium used to culture and observe the morphological characteristics of actinomycetes. With the addition of appropriate amounts of antimicrobial agents (e.g., various antibiotics, phenols, etc.), it can be used to isolate a variety of actinomycetes. The main feature of this synthetic medium is that it contains a variety of inorganic salts with known chemical compositions, which may interact with each other and produce precipitation, such as phosphate and magnesium salts in Gao's No. I medium, which are prone to precipitation when mixed with each other. Therefore, when mixing the components of the medium, it is usually necessary to dissolve the components in the order of the formula, and it is even sometimes necessary to sterilize the two or more components separately, and then mix them according to the proportions when they are used. In addition, some synthetic media also need to be sterilized, and then mixed in proportion when used. In addition, some synthetic culture media have to be supplemented with trace elements, for example, the amount of FeS04-7H2O in Gao's No. I culture media is only 0.001%, so it is necessary to formulate a high concentration of FeS04-7H2O storage solution in advance, and then add a certain amount to the culture media according to the need.

Soluble starch, KNO3, NaCl, agar, K2HPO4-3H2O, MgS04-7H2O, FeS04-7H2O, 1 mol/L NaOH, 1 mol/L HCl.2. Instruments or other paraphernalia

Test tubes, triangular flasks, beakers, measuring cylinders, glass rods, medium dispensers, scales, crocks, autoclave, pH test paper (pH 5.0-9.0), cotton, kraft paper, markers, twine or rubber bands, gauze, etc.
III. Operational steps
1. Weighing and solubilization
(1) According to the formula, first weigh the soluble soluble powder and put it into a small beaker, and use a small amount of cold water to make the starch into a paste, and then add less than the required amount of boiling water, and continue to heat it, so that the soluble starch is completely dissolved.

(2) Then weigh the other ingredients and dissolve them one by one.

(3) For the trace component FeS04-7H2O can be prepared into a high concentration of storage solution in proportion to the conversion and then added, the method is to add 1 g of FeS04-7H2O in 100 ml of water to make 0.01 g/ml, and then add 1 ml of 0.01 g/ml of storage solution to 1000 ml of culture medium.

(4) After all the drugs are completely dissolved, replenish the water to the total volume required.
