Summary

This is a method for the specific detection of IL-12 heterodimers. Since IL-12 p4 0 monomer cannot be detected, this method is able to detect IL-12 p7 5 heterodimer in the presence of excess IL-12 p4 0 subunit. Author: J.E. BASIC PROTOCOL E L I S A DETECTION OF HUMAN AND MOUSE IL-12 p7 5 ISODIMERS Materials

9 . 吸去或倒去孔中液体，按照步骤3 洗 板 5 次 。 10• 用 检 测 缓 冲 液 稀 释 H RP偶 联 抗 体 4D6 至 250ng/m l或 H R P 结 合 抗 体 5C3 至 500ng/ml。 每孔加100/xl稀释抗体，室温孵育2h。 11•按照步骤3 洗 板 5 次 ，然后每孔中加入100W 发色底物。室温孵育10〜15min。 12. 每孔中加入lOOyl终止液，用 E L I S A 读板仪根据H R P 底物对应的波长读板。 13. 绘 制 标 准 曲 线 （吸光度比pg/mlIL-12)，并根据标准曲线计算未知样品中IL-12的 浓 度 （见 单 元 5. 2 中细胞因子E L I S A 计算结果）

Options ELSA for IL-12 p4 0 Subunit in Human and Mouse IL-2异二聚体产物常常伴随着IL-12p4 0 亚 基 产 物 的 产 生 （通常> 5 倍 ）。基本方案 里采用能识别IL-12 P4 0 异二聚体和游离形式的亚基的包被抗体用来检测这两种形式的 产物。针 对 人 IL-12,用小鼠抗人IL-12p4 0 亚基单克隆抗体2-3A 1 ( 由 R .
