In vitro antimicrobial effect of traditional Chinese medicines in paper slice experiments

Materials and Instruments

Plain agar plates Dry drug sheets Inoculation rings Small forceps Staphylococcus aureus slants

Move

1. Pick appropriate amount of Staphylococcus aureus slant culture with sterilized inoculation ring, start from one side of the agar plate, inoculate the bacteria on the plate with dense scratching method, rotate the plate while scratching until the whole plate is uniformly scratched all over, and then cauterize the inoculation ring to sterilize it.

2. Use the cauterized tweezers to pick up 1~5 pieces of paper with quantitative drug absorption, place them evenly on the surface of the plate, and slightly press the paper to make it in good contact with the culture medium.

3. After 18 to 24 hours of incubation at 37℃, observe whether there is a circular area without bacterial growth around the paper pieces (inhibition circle), and measure the diameter of the inhibition circle to determine whether the bacteria are sensitive to the drug and the size of the drug's ability to inhibit bacteria.
