Morphological observation experiment of Trichomonas vaginalis

Principle

Trichomonas vaginalis is a parasite of the vagina and urethra in women or the prostate and urethra in men, and can cause trichomoniasis or urethritis. The life history of the parasite consists of a trophozoite stage without an encysted stage. Typical trophozoites are pear-shaped or oval, and when in poor condition or aging, the body becomes rounded, with a large number of refractive particles in the cytoplasm, or even with vacuoles. The size of the trophozoites varied greatly with the source of the species and the time of division, generally 10-15??m wide, 7-32??m long, colorless and transparent, with refractive properties. The anterior end of the body has four equal lengths of the anterior flagellum, the fifth flagellum backward, along the undulating membrane (undulating membrane) outer edge of the inner side of the wavy extension, and the outer edge of the undulating membrane is as long as the length of the outer edge, no free edge. The undulating membrane is a double membrane structure formed by the outward elevation of one side of the body of the worm, with a smooth surface, and it is the motile organelle of the body of the worm. The axostyle extends from the anterior end to the posterior end of the body, runs through the body, and extends out at the posterior end of the body, with the tip of the tip, attached, often with vaginal epithelial cells or debris, etc. The nucleus is located in the anterior 1/3 of the body, without a free edge. The nucleus is located in the anterior 1/3, an oval vesicular nucleus, the upper edge of the nucleus has 5 basal bodies arranged in a cup shape, from which flagella are emitted. Inside the cytoplasm, there are deeply stained granules arranged parallel along the axial column, which are hydrogenosome, a unique enzyme system of the worm. The body of the worm is soft and versatile, highly mobile, without directional movement, and the fluctuating membrane makes the worm rotate, while the anterior flagellum acts as a propelling force (Fig. 1, Fig. 2). 1. Trichomonas vaginalis stained specimens

The body of Trichomonas vaginalis was mostly pear-shaped, 7-32 um long, with four anterior flagella, short fluctuating membrane, only 1/2 of the length of the body, no cell mouth, and the axonemal column was elongated and protruded backward out of the body. The nucleus was located in the anterior part of the body.

2. Trichomonas vaginalis live specimen

Take the vaginal secretion of patients with trichomonas vaginitis for hanging drop method or direct smear method, observe the live trichomonas under low or high magnification microscope, pay attention to the observation of trichomonas in the low magnification microscope special jumping rotation, in the high magnification microscope can be seen in the flagellum and fluctuating membrane of the activities. Because of the great mobility of the worm, it is not easy to observe in detail, and a drop of serum diluted 1:10 can be added to the smear specimen to hinder the strong movement of Trichomonas and facilitate observation. Large polygonal vaginal epithelial cells and small round white blood cells as well as pus cells gathered in piles can also be found in vaginal secretion smears and should be recognized.
