G K 1.5 ( A T C C 号 TIB207 )、 53-6.72 ( TIB105)、 M 5/114. 15.2 ( TIB120)、 Jlld 2 (TIB183; PharMingen; 单克隆抗体配方见附录1) 单克隆抗体M A R 18. 5 (TIB216; PharMingen) 浓 度 lmg/ml (单克隆抗体配方见 附 录 1) V 含酚红的 H B S S (Life Technologies) /10% (V /V ) FBS V 低 Tox-M 兔补体，无菌复溶 淋巴细胞M 密度梯度分离液（Accurate Chemical and Sciendfic)，室温 V R P M I - I O 完全培养基 F F T C 标记的抗C D 3 抗 体 和 P E 标记的P K 136抗 体 （Pharmingen) 50m l 和 15m l 带盖聚丙烯锥底离心管 Beckman C S -6R 离 心 机 （或同类离心机） 血清学吸管 注 ：如果细胞用于继续培养，所有试剂和材料必须保证无菌。 1 . 处死小鼠，无菌取脾脏。 2 . 制 备 脾 脏 细 胞 悬 液 （单 元 2.1)，置 于 50m l 离心管中，避免将纤维性杂质移入离心 管 。 200g 离 心 5m i n 以 洗 涤 细 胞 。采 用 单 元 2 . 1 的 方 法 裂 解 红 细 胞 ，或使用 T W N H 4C l 裂解缓冲液。每个脾脏的细胞用Iml H B S S / 3 % F B S 混悬。 3 . 用台盼蓝拒染法计数活细胞（附 录 3C )。用 H B S S / 3 % F B S 混 悬细胞至5 X 107 个细 胞/ml。置于冰上保存。 4 . 制备下述单克隆抗体的I O X 混 合 溶 液 （10X 每种抗体的饱和浓度）： G K 1.5 (抗 C D 4) 35-6.72 (抗 C D 8) M 5/114. 15. 2 (抗 I-A 和 I-E d’k) Jlld.2 (抗 C D 24) 5 . 每 0.9m l 细胞悬液加入0.1m l 单克隆抗体混合液， 4°C孵 育 30min。 6 . 用 冰 的 H B S S / 3 % F B S 洗 细 胞 2 遍 ， 4°C ， 200g 离 心 5min，弃上清。用 IOml冰的 H B S S / 3 % F B S 混悬细胞再洗一遍。将 细 胞 用 H B S S / 1 0 % F B S 混悬至浓度为5 X 107 个细胞/m l 。 7 . 加 入 10Mg/m l 抗 大 鼠 ic (M A R 18.5; lmg/m l 储存液）和 低 Tox-M 兔补体至细胞悬 液 使 其 达 到 最 佳 终 浓 度 （通 常 1 : 6 , 但需要根据经验确定）。 37°C 孵 育 45min，每 15m i n 振摇一次。 8•用 H B S S /10% F B S 洗 一 遍 （步骤 6)。 9 . 用 5m l H B S S / 1 0 % F B S 混悬细胞。取少量细胞悬液采用台盼蓝拒染法计数活细胞 (附录3 0 , 以确定补体裂解效果。 10. 将 5m l 室温的淋巴细胞M 密度梯度分离液加入到一个15m l 离心管中。将 5m l 细胞 悬液轻轻加在分离液上。室温， 200g 离 心 20min，使离心速度缓慢下降。 11. 用吸管从界面小心吸取液体，放 入 另 一 个 15m l 离心管中。必要时可将界面放在白 色背景前以便看清。用 R P M I -I O 完全培养基洗2 遍 （方法同 步 骤 6)。台盼蓝染色 计数活细胞，备用。

12. To test for cellular purity, anti-CD3-FITC and PK136-PE (anti-NK1.1) antibodies at a final concentration of lOug/ml can be added and analyzed by flow-through analysis (Chapter IV).
