Peripheral heparin anticoagulated blood (Appendix 3G), tonsils, spleen cells or bone marrow

lmg/m l Cyclin A, 100% (V/V) ethanol, stored at 20°C.

Beckman 4°C centrifuge and JS4.2 rotor (or alternative)

0.45um filter

50m l conical bottom centrifuge tube

25c m 2 cell culture flasks

1•将I X l O 6个细胞/m l 对 数 生 长 期 的 B 95-8细 胞 接 种 到 R P M I -I O 完全培养基中。置 37°C ， 5 % C 0 2细胞培养箱中培养3d 。确保活细胞> 9 0 % (附录3 0 。 2 , 细胞经 4°C ， 300g 离 心 IOmin后 ，分 离 含 E B V 的 上 清 O l O 2〜IO3转化单位/ml)。 上 清 经 0.45/m i滤器过滤除菌，分装 ， 一 130°C 可 保 存 1 年或更长时间。最好对病毒 上清进行滴定检测（Miller and Lipman， 1973)0 3•用P B S 将 约 15m l 外周肝素抗凝血进行I : 2 稀 释 。使用扁桃体或脾脏细胞时，需制 备 单 细 胞 悬 液 （单 元 7. 8)，用 R P M I -I O 完全培养基调整浓度为5X I O 6〜I X I O 7个细 胞/m l 。使用骨髓细胞时，用 R P M I -I O 完全培养基将骨髓细胞1:20 (V A O 稀释。 4 . 在 50m l 锥底离心管中，将 12m l 稀 释 后 的 血 液 样 品 （或细胞悬液、骨髓）轻轻叠加 到 12ml Ficoll-Hypaque分离液上。室温， 1500g 离 心 8min，吸取界面层细胞，加入
50m l 锥底离心管中。 5•加入适量P B S ，室温， 300g 离 心 15min，弃上清。用 H B S S 重 悬 细 胞 ， 300g 离心 lOmin，弃上清。细 胞 用 H B S S 再 洗 涤 1 遍 ，加 入 2〜5ml R P M I -10完全培养基重悬 细 胞 并 计 数 （附 录 3A )。 6 . 在 50m l 锥底离心管中加入IO7个单个核细胞，总体积2. 5m l 。然后加入2.5m l 步 骤 2 中的病毒上清。 37°C 水 浴 2h 。 7 . 加 入 5m l 含 lMg/m l 环 胞 素 A 的 R P M I -I O 完全培养基。然 后 将 细 胞 悬 液 （约 IOml) 移 入 25cm2细胞培养瓶中。置 37°C ， 5 % C 〇 2细胞培养箱中培养3 周 。培养期间在相 差显微镜下观察B 细 胞 转 化 情 况 （3 周左右时，细胞体积变大，聚集形成集落）。 8. 3 周后，混匀细胞，取 5m l细胞悬液加入新的25cm2细胞培养瓶中。每瓶再加入 5ml R PM I-IO 完全培养基，培 养 1 周 。此时可将细胞一 130°C冻 存 ，或 每 周 用 RPMI-IO 完全培养基将细胞按1 : 3 传代培养
