Is calcium ion-containing HBSS an option for the preparation of mouse tumor single cell suspensions?

It is not recommended because calcium and magnesium ions can inhibit enzyme activity. Although most enzyme reagents on the market today contain EDTA to chelate calcium and magnesium ions, it is recommended that you use calcium and magnesium ion free HBSS. It is also recommended that you use calcium and magnesium ion free HBSS if the subsequent assay contains indicators related to calcium ions, e.g., calcium ion endocytosis. If the subsequent assay contains indicators that require stimulation of activation, it is also important to use an enzyme that does not contain EDTA, e.g., IFNγ produced by activated T-cells (because EDTA chelates calcium ions, which are an important micronutrient in the activation process of T-cells).
