Ultraviolet sterilization test

Summary

Ultraviolet sterilization is carried out with ultraviolet lamps, the wavelength of 200 ~ 300 nm of ultraviolet light have the ability to sterilize, of which 260 nm of the strongest bactericidal force. It is mainly used to sterilize (1) the surface of objects and (2) the air in surgical rooms.

Principle

As radiation can make the oxygen in the air ionized into [O], and then oxidize O2 to generate ozone (O3) or make water (H2O) oxidized to generate hydrogen peroxide (H2O2), O3 and H2O2 have bactericidal effect. UV penetration is not large, so it is only suitable for sterilizing the air and the surface of objects in the sterile room, inoculation box, operating room. UV lamp from the irradiated object to not more than 1.2 m is appropriate.

1. Ultraviolet radiation alone
(1) Turn on the UV lamp switch in the aseptic chamber or in the inoculation box, irradiate for 30 minutes, and turn the switch off.
(2) Open the lid of the beef paste peptone flat dish for 15 minutes, then cover the dish and incubate at 37°C for 24 hours. A total of three sets were made.
(3) Check the number of colonies growing on each flat dish. If there are no more than 4, it means that the sterilization effect is good, otherwise, it is necessary to extend the irradiation time or strengthen other measures at the same time.
2. Chemical disinfectants in combination with ultraviolet irradiation

(1) In a sterile room, spray a 3~5% solution of carbolic acid and then irradiate with ultraviolet light for 15 minutes.
(2) Scrub the table tops and stools in the sterile room with 2~3% Lysol and then turn on the UV lamp for 15 minutes.
(3) Check the sterilization effect.
Since UV rays have a damaging effect on the conjunctiva and optic nerve of the eye, and an irritating effect on the skin, you should not look directly at UV light, let alone work under UV light.

Common Problems
