Dermatol Sulfate (DS), also mistakenly referred to as chondroitin sulfate B, is a class of sulfated GAGs composed of disaccharide units (GlcUAa1-3GalNAc) composed of iduronic acid (IdoUA) and N-acetylgalactosamine (GalNAc) repeatedly linked by β -1,4-glycosidic bonds. From the perspective of composition and structure, DS is a type of GAGs that is completely different from CS. However, from the perspective of biosynthesis, DS is a product of the transformation of glucuronic acid residues (GlcUA) in the sugar chain into IdoUA residues under the action of C5 differential isomerase during CS synthesis. Therefore, CS and DS often coexist in the same sugar chain and are called CS-DS hybrid chains. Numerous studies have shown that the DS region in the CS-DS sugar chain typically plays a critical role in interactions with proteins and participates in various important physiological and pathological processes along with CS. At present, commercial DS mainly separates animal skin or heparin by-products, but due to its high similarity with HS, CS, and HA in physicochemical properties, it is difficult to prepare high-purity DS.
