Descripción general

Cholesterol, also known as cholesterin, is a derivative of cyclopentanoperhydrophenanthrene. It is widely present in animal bodies, with the highest concentrations found in the brain and nerve tissues. It is also relatively abundant in the kidneys, spleen, skin, liver, and bile. The enzymatic method for determining Total Cholesterol (TC) is commonly used in biochemical testing. In blood, approximately one-third of cholesterol exists as free cholesterol, while two-thirds is in the form of cholesterol esters bound to fatty acids. These cholesterol esters are hydrolyzed into free cholesterol by cholesterol esterase (CEH). The free cholesterol is then oxidized by cholesterol oxidase (COD) to form cholesterone, producing hydrogen peroxide in the process. Subsequently, in the presence of peroxidase (POD), hydrogen peroxide catalyzes the reaction between 4-aminoantipyrine and phenol (collectively referred to as PAP), generating a red quinoneimine dye (Trinder reaction). When CEH is absent, cholesterol esters cannot be hydrolyzed, allowing only the measurement of free cholesterol content. The absorbance is measured using a spectrophotometer at wavelengths between 500 and 520 nm. This assay kit is primarily designed for the quantitative determination of free cholesterol in samples such as human or animal serum, plasma, cerebrospinal fluid, cells, and tissues. It is intended for research use only and is not suitable for clinical diagnosis or other purposes.
