N-Acetylglucosamine 1-phosphate uridyltransferase (PmGlmU), EC 2.3.1.157, derived from Pasteurella multocida. This enzyme is used to catalyze the formation of UDP‑GlcNAc from GlcNAc‑1‑P, working in conjunction with PmPPA and NahK to activate GlcNAc. The product is recombinantly expressed in E. coli and exhibits high activity.
Reaction
GlcNAc‑1‑P + UTP → UDP‑GlcNAc + PPi
Protocol
Add substrates to the reaction system according to the desired composition. Include 5 mM MgCl₂ and, if desired, a buffer salt such as Tris‑HCl. Adjust the pH to 7.5–8.5, then add an appropriate amount of PmGlmU (depending on the reaction requirements).
Mix thoroughly and incubate the sample at 37 °C for 30–60 min. The recommended reaction buffer is Tris‑HCl (pH 8.0), but other common biological buffers such as Tris or PBS are also suitable.
Note: When scaling up the system, the incubation time or enzyme amount should be adjusted according to actual conditions; low‑speed shaking may be used to ensure proper mixing.
