Descripción general

Malachite Green, also known as Aniline Green, is a synthetic dye with CAS No. 569-64-2 and a molecular weight of 364.92. Glycerin-Malachite Green Staining Solution is mainly composed of glycerin, malachite green, and deionized water, and can be used to examine various helminth eggs in feces. This reagent is for research use only and is not suitable for clinical diagnosis or other purposes.

Operating Steps (For Reference Only)

  1. Prepare glass paper strips: Soak the glass paper in Glycerin-Malachite Green Staining Solution for at least 24 hours.

  2. Place a small amount of fecal sample on newspaper or a small paper sheet. Press a filter screen onto the fecal specimen so that part of the sample passes through and accumulates on the screen.

  3. Use a spatula to scrape across the filter screen to collect the sieved fecal sample.

  4. Place a perforated plate in the center of a glass slide. Fill the hole with the fecal sample using the spatula, and scrape the plate surface with the edge of the spatula to remove excess sample around the hole.

  5. Carefully remove the plate, leaving a small cylindrical fecal sample on the slide.

  6. Take a glass paper strip soaked in the staining solution and cover the fecal sample with it.

  7. Flip the slide and press the fecal sample against another slide or a smooth, hard surface to spread the specimen evenly between the slide and the glass paper strip. After clarification, the text on this instruction sheet should be readable through the smear.

  8. Gently slide and remove the top slide from the side, avoiding separation from or lifting of the glass paper strip. Place the slide on the bench with the glass paper strip facing upward. For most eggs (except hookworm eggs), allow the slide to stand at room temperature for one to several hours until the sample clears.

  9. Examine the smear by moving up and down or side to side. Report the count of each type of egg found, then multiply by the appropriate factor to obtain the number of eggs per gram of feces:

    • For a 20 mg plate, multiply by 50.

    • For a 41.7 mg plate, multiply by 24.

    • For a 50 mg plate, multiply by 20.


Precautions

  1. The spatula and filter screen can be reused after careful cleaning.

  2. For dry fecal samples, the covering glass paper strip must be well moistened. For soft stools, the glass paper strip can be slightly less moist. In dry climates, excess glycerin can only delay, not prevent, drying of the fecal sample.

  3. In preparations made by this method:

    • Ascaris and Trichuris eggs can be preserved for a long time.

    • Schistosome eggs can be preserved for several months.

    • Hookworm eggs often become unobservable 30–60 minutes after preparation.

  4. This staining method requires specific fecal film thickness and clarification time. If the film is too thick and clarification time is too short, eggs may be difficult to detect. If clarification time is too long, eggs may deform and become hard to recognize. For examining hookworm eggs, the clarification time should be controlled within 30 minutes.
