Materials	- Catabolic enzyme: Immobilized Cephalosporin C Acylase (≥130 U/g wet weight, 60% water content) - CPC-Na - NaHCO₃ - Water - 1.0N NaOH - HPLC - Chromatography column (Symmetry C18, 5μm, 250mm×4.6mm)
Condition	- pH: 8.3 - Temperature: 13℃ - CPC - Na: 75mM - Enzyme: 10 U/ml (with 130ml)- Reaction solution: Water
Step	1. Remove the water after washing immobilized enzyme with water more than three times. 2. Dissolve 75mM CPC-Na and 0.3% NaHCO₃ in 130ml. 3. Leave the substrate in gradient cylinder for 10min at 20℃. 4. Pour the catabolic enzyme (Immobilized Cephalosporin C Acylase, 6.5 U/ml) in conversion reactor and measure time with pouring substrates. 5. Adjust pH to 8.3 with 1.0N NaOH. 6. Extract 0.1ml reaction solution (supernatant) and dilute with water in 100ml flask. 7. Measure the yield of 7-ACA through peak area in HPLC detector.
