Incubation method for fecal testing

Principle

The eggs are incubated and under certain conditions the larvae can be observed with the naked eye in a specific pattern of activity. This method has a high degree of specificity and includes the miracidium hatching method, which is primarily used to detect schistosomes, and the culture method for hookworm larvae.

(i) Trichinella incubation method

1、Use:Applicable to fecal examination of patients with early schistosomiasis.

2、Method

(1) Take 20-30g of feces, processed by natural sedimentation method.

(2) Pour the precipitate into 250~300ml glass triangular flask, add water or overnight dechlorinated tap water to the mouth of the flask, and place it in a warm box at 25℃~30℃ for incubation.

(3) After 4 to 6 hours of the first observation, take out the triangular flask placed in front of the dark-colored background material, using the naked eye or magnifying glass to observe the mouth of the flask near the water surface, white dotted trichomes can be seen to move in a straight line. If negative, observe once more after 12 to 24 hours.

(ii) Cultivation of larvae

1、Use:Applicable to the fecal examination of patients with hookworm disease.

2、Method

(1) Take a small test tube containing 1~2ml of cold boiled water.

(2) Cut the filter paper into a "T"-shaped strip as wide as the diameter of the small test tube and slightly shorter than the length of the tube.

(3) Use a bamboo stick or toothpick to pick about 0.2g (soybean size) of feces, evenly spread on the upper 2/3 of the filter paper.

(4) Insert the feces-coated strip of filter paper into a small test tube and immerse the lower end of the strip into cold boiled water without touching the bottom of the tube to keep the filter paper moist (care must be taken to ensure that the feces on the strip does not get mixed into the water).

(5) In the upper end of the test tube from the mouth of the 3cm labeling, standing in the test tube rack, placed in 25 ℃ ~ 35 ℃ room temperature or incubator culture for 3 to 7 days.

(6) Add cold boiled water along the wall of the tube with a rubber-tipped pipette every day to replenish the water lost through evaporation.

(7) Hookworm eggs in feces are incubated on moist filter paper strips for 3 to 7 days, and the hookworm larvae that hatch enter the water at the bottom of the test tube. When examined, the strip of filter paper in the test tube is first removed, and then observed with the naked eye, the silver-white wriggling larvae of the hookworms are visible in the water at the bottom of the tube.
