Wheat seeds: first soaked for 15 hours, i.e. it is appropriate to show white soon after sucking up.
Naphthalene acetic acid or sodium naphthalene acetate 100pp M solution: NAA should be dissolved first in a small amount of 95% ethanol, and finally fixed to the required volume.
5% bleach: 5 g of bleach is fixed to 100 ml .
II. Experimental Steps
Number the washed and dried petri dishes sequentially. Add 10 ppM naphthalene acetic acid solution 10 ml into ① and add 1 ml from ① into ②, and add 9 ml distilled water into ②, mix well, then it will become 1 ppM naphthalene acetic acid solution, and then continue to dilute it to ⑤, then it will be 10, 1.0, 0.1, 0.01, 0.001 ppM concentration, and then finally 1.0 ml was sucked out from ⑤, and then it was disposed off. The last 1.0 ml of ⑤ was aspirated and discarded. 9 ml of distilled water was added to the ⑥ dish as control.
Select 120 pure wheat seeds, put them in a small beaker, add appropriate amount of 0.1% mercuric chloride or 5% bleach to sterilize them for 10 or 20 minutes, first rinse them with tap water for several times, then immerse them for 15 hours, then rinse them with distilled water for two times, and finally rinse them with redistilled water for two times, use filter paper to suck out the water adhering to the seeds, and put a piece of filter paper in each Petri dish, and put the 20 immerged seeds neatly on the filter paper, so that the seed embryo is facing the filter paper. The seeds were placed on the filter paper so that the embryos were facing the filter paper, and the petri dishes were put into a 25℃ incubator after being covered with a lid. After three days, the average number of roots, average root length and average shoot length in different treatments were determined and recorded in the table below.
