Indirect method for the detection of anti-human globulin test

Summary

The purpose of the indirect test is to check for the presence of free incomplete antibodies in the serum. First, Rh(D)-positive O-type (or the same type as the examined ABO) normal human erythrocytes are used to adsorb the free incomplete antibodies (i.e., called sensitized) present in the serum. Sensitized erythrocytes are washed with saline and then added with anti-human globulin serum, and agglutination occurs, i.e., anti-human globulin is positive in the indirect test. This experiment is derived from the laboratory instruction of Mudanjiang Medical College undergraduate 5-year testing program.

Principle

The purpose of the indirect test is to check for the presence of free incomplete antibodies in the serum. Rh(D)-positive O-type (or the same type as the ABO being examined) normal human erythrocytes are first used to adsorb the free incomplete antibodies present in the serum (i.e., called sensitization) The sensitized erythrocytes are washed in saline and then added to the anti-human globulin serum, and agglutination occurs, which is considered to be a positive indirect test for anti-human globulin.

Method I:

1. Take 0.5-1.0 milliliters of the examined blood cyanide and add an equal amount of 10% mixed O-erythrocyte suspension. Mix to 37 ℃ water bath for one hour.

2. Wash three times with large amounts of saline, and then make a 10% red blood cell suspension.

3. Report the operation according to one of the direct test methods.

Method II:

1. Take four test tubes and proceed according to the table.

2. After mixing, sensitize in a 37°C water bath for 1 hour.

3. Centrifuge to remove the supernatant, wash three times with saline, and then prepare 5% erythrocyte suspension with saline in each tube.

4. Take 4 small test tubes and operate according to the table:


5. After mixing, leave at room temperature for 30 minutes and observe the results.

Indirect test is a method of checking the presence of free incomplete antibodies in the patient's serum. Patients with hemolytic anemia caused by blood group incompatibility are mostly positive, and the mother's serum of neonates with hemolytic disease can be positive.
